Signal transducer and activator of transcription 5A (Stat5a) and Stat5b, which share 96% homology, undergo receptor tyrosine kinase or G protein-coupled receptor-dependent phosphorylation in response to cytokines or growth factors, and then form homo- or heterodimers that translocate to the nucleus, where they initiate transcription. Activation of Stat5a via IL-2, IL-3, IL-7/ GM-CSF, erythropoietin, thrombopoietin and growth hormones influences proliferation, differentiation and apoptosis in lymphohematopoietic cells. Calretinin (also known as Calbindin 2) is a member of the troponin C superfamily. Proteins in this family have 6 EF-hand domains which bind to calcium. Calretinin is a vitamin D-dependent calcium-binding protein involved in calcium signaling. It is present in subsets of neurons throughout the brain and spinal chord, including sensory ganglia. Antibodies to calretinin are useful in differentiating mesothelioma from adenocarcinomas of the lung. They can also aid in differentiating adrenal cortical neoplasms from pheochromocytomas. SOX2 is required for stem cell maintenance in the central nervous system, and it also regulates gene expression in the stomach. SOX2 is necessary for regulating multiple transcription factors that affect Oct 3/4 expression. An essential function of SOX2 is to stabilize embryonic stem cells in a pluripotent state by maintaining the requisite level of Oct 3/4 expression. The NF-E2 DNA binding protein is composed of two subunits, p45 and MafK, and it regulates expression of globin genes in developing erythroid cells through interaction with Maf recognition elements (MAREs). A family of NF-E2 related proteins, which are collectively known as the Cap 'n' collar (CNC) family and include Nrf1 (also designated TCF11), Nrf2 and Nrf3, are bZIP transcription factors that heterodimerize with Maf proteins to bind MARE sequences. Mutations in DNA mismatch repair genes are associated with hereditary nonpolyposis colorectal cancer (HNPCC). The v-Myc oncogene, initially identified in the MC29 avian retrovirus, causes myelocytomas, carcinomas, sarcomas and lymphomas, and belongs to a family of oncogenes conserved throughout evolution. In humans, the family consists of five genes: c-Myc, N-Myc, R-Myc, L-Myc and B-Myc. Amplification of the N-Myc gene has been found in human neuroblastomas and cell lines. Its amplification correlates well with the stage of neuroblastoma disease. Immunological studies have shown that the human N-Myc gene encodes a nuclear phosphoprotein that exhibits relatively short (30 min) half life in vivo. This antibody recognizes an epitope encoded by exon v3 on the variant portion of human CD44. The CD44 molecule belongs to a family of cellular adhesion molecules found on a wide range of normal and malignant cells in epithelial, mesothelial and hemopoiesis tissues. CD44 is a single gene with 20 exons, of which 10 are normally expressed to encode the basic CD44 (H-CAM) molecule. The additional 10 exons (v1 to v10) are only expressed by alternative splicing of the nuclear RNA. The expression of specific cell adhesion molecule CD44 splice variants has been reported to be associated with metastasis in certain human malignancies.
