Each vial contains 10 mg/mL BSA (free of IgG and protease) as a stabilizer and 0.01% sodium azide as a preservative, after reconstitution. Concentration is 1.0 mg/mL when reconstituted as directed. Biotium Cytokeratin 5/6/18(LP34), CF568 conjugate, 0.1mg/mL
Recognizes polypeptides of 58 kDa, 56 kDa, and 45 kDa, identified as cytokeratin 5, 6, and 18 respectively. It shows no reaction with keratin 1, 8, or 19. This MAb shows a broad pattern of reactivity with epithelial tissues, from simple glandular epithelia to stratified squamous epithelia. Epithelial cells are labeled whether they are ectodermal, mesodermal, or endodermal in origin. This MAb is useful in detecting cells of epithelial origin in mixed tumors, in effusions, in bone marrow samples, or in tissue culture cells.Primary antibodies are available purified, or with a selection of fluorescent CF Dyes and other labels. CF Dyes offer exceptional brightness and photostability. Cytokeratin 8 (CK8) belongs to the type II (or B or basic) subfamily of high molecular weight cytokeratins and exists in combination with cytokeratin 18 (CK18). This MAb cocktail recognizes all simple epithelia including glandular epithelium, for example thyroid, female breast, gastrointestinal tract, respiratory tract, and urogenital tract including transitional epithelium. All adenocarcinomas and most squamous carcinomas are positive but keratinizing squamous carcinomas are usually negative. This antibody is useful in demonstrating the presence of Paget cells; there is very little keratin 18 in the normal epidermis so only Paget cells are stained. Cytokeratin 13 (KRT13) is the major acidic keratin, which together with KRT4, its basic partner, is expressed in the suprabasal layers of non-cornified stratified epithelia including tongue mucosa, esophagus, anal canal epithelium, tracheal epithelium, uterine cervix, and urothelium. Defects in the KRT13 gene are a cause of white sponge nevus of cannon (WSN), a rare autosomal dominant disorder, which predominantly affects non-cornified stratified squamous epithelia and is characterized by the presence of soft, white and spongy plaques in the oral mucosa. KRT13 has been used as a marker for non-keratinized squamous epithelium. It is also expressed in various squamous metaplasia, but it is down regulated in squamous dysplasia and squamous carcinoma. This MAb recognizes a protein of 56 kDa, identified as cytokeratin 6A (KRT6A). In humans, multiple isoforms of Cytokeratin 6 (6A-6F), encoded by several highly homologous genes, have distinct tissue expression patterns. Cytokeratin 6A is the dominant form in epithelial tissue. Cytokeratin 6 and 16 are expressed in keratinocytes, which are undergoing rapid turnover in the suprabasal region (also known as hyper-proliferation-related keratins). Cytokeratin 6 is found in hair follicles, suprabasal cells of a variety of internal stratified epithelia, in epidermis, in both normal and hyper-proliferative situations. Epidermal injury results in activation of keratinocytes, which express KRT6 and KRT16. KRT6 is strongly expressed in about 75% of head and neck squamous cell carcinomas. This MAb reacts with a wide variety of simple epithelia. It does not react with stratified squamous epithelia. It reacts with epithelial tumors of the gastrointestinal tract, lung, breast, pancreas, ovary, and thyroid. Cytokeratin 18, which belongs to the type A (acidic) subfamily of low molecular weight keratins, exists in combination with cytokeratin 8. It was reported that tissues from gastrointestinal tract are positive for both cytokeratin 8 and 18 but do not contain cytokeratin 14.
